Its goal is to completely map and sequence all of the genetic material that makes us human

When it is done, we will have a new and profoundly powerful tool to help us to unravel the mysteries of how the human body grows and functions.

The cells in our bodies each contain a master program which controls how and when they develop and how they should function. This information is organised in units called genes, which are arrayed, one after the other along long polymers called chromosomes. We have 46 chromosomes, arranged in pairs kept in the nucleus of most cells. The chromosomes are made of deoxyribonucleic acid, or DNA

It is comprised of just four building blocks, or residues, strung together in enormously long strings. The residues combine to make our genes, and our genes string together to make our chromosomes.

A genetic linkage map shows the relative locations of specific DNA markers along the chromosome. Any inherited physical or molecular characteristic that differs among individuals and is easily detectable in the laboratory is a potential genetic marker. Markers can be expressed DNA regions (genes) or DNA segments that have no known coding function but whose inheritance pattern can be followed. DNA sequence differences are especially useful markers because they are plentiful and easy to characterize precisely. Cells have a second type of nucleic acid - RNA (Ribonucleic Acid) which can also carry genetic information

The messenger RNA (mRNA) carries genetic information from the DNA to the ribosomes

where it is translated into a protein. mRNA is synthesized in the nucleus based on a single DNA strand, using the RNA polymerase enzyme. mRNA is transcribed from a DNA strand only in locations called open reading frames

In eukaryotes, the mRNA is formed of coding and non-coding regions. Coding regions are the regions used to carry real genetic information. Non coding regions do not carry such information (see below).

The coding regions are called exons, since they are able to leave the nucleus and reach the Ribosome. The non-coding regions are called introns and never leave the nucleus.

Physical maps describe the chemical characteristics of the DNA molecule itself.

chromosomal mapping can be used to locate genetic markers defined by traits observable only in whole organisms. Because chromosomal maps are based on estimates of physical distance, they are considered to be physical maps. The number of base pairs within a band can only be estimated.

New genes can be located by combining genetic and physical map information for a region. The genetic map basically describes gene order. Rough information about gene location is sometimes available also, but these data must be used with caution because recombination is not equally likely at all places on the chromosome. Thus the genetic map, compared to the physical map, stretches in some places and compresses in others, as though it were drawn on a rubber band.

Technological improvements now make possible the cloning of large DNA pieces, using artificially constructed chromosome vectors that carry human DNA fragments as large as 1 Mb. These vectors are maintained in yeast cells as artificial chromosomes (YACs). Yeast Artificial Chromosomes (YACs), or pieces of human DNA carried in yeast, in order to complete the map. The investigators compared 16 of these YACs and used the common, overlapping regions to determine the correct order of the DNA and to locate a series of markers along the chromosome.

YAC methodology drastically reduces the number of clones to be ordered; many YACs span entire human genes. A more detailed map of a large YAC insert can be produced by subcloning, a process in which fragments of the original insert are cloned into smaller- insert vectors. Because some YAC regions are unstable, large- capacity bacterial vectors (i.e., those that can accommodate large inserts) are also being developed.

Bacterial Artificial Chromosomes (BACs) are the most broadly used DNA clone resource for large genome sequencing programs. Sequence reads at the ends of cloned DNAs mark the boundaries of DNA contained within, and are called sequence tag connectors (STCs

The BACs representing a genome can together serve as a scaffold on which much shorter DNA sequence assemblies can be located.

The physical maps of the Y chromosome that weve made should make it much easier to explore the biology of the chromosome that to this time has been one of the most mysterious, said Page. The Y chromosome is particularly mysterious because it is difficult to investigate through family inheritance studies, he said. I think the biology of the Y chromosome can only be explored from the DNA level up.

scientists has found that a specific defect in the male sex chromosome (the Y chromosome) may be responsible for 1 percent of cases of azoospermia, the complete inability to make sperm and the most severe form of male infertility. The study is one of the first to demonstrate that genetic defects can sometimes explain infertility in otherwise healthy couples and could lead to a better understanding of the molecular mechanisms required to make healthy sperm

Fairness in the use of genetic information by insurers, employers, courts, schools, adoption agencies, and the military, among others.

Who should have access to personal genetic information, and how will it be used?

Psychological impact and stigmatization due to an individuals genetic differences.

How does personal genetic information affect an individual and societys perceptions of that individual?

How does genomic information affect members of minority communities

Reproductive issues including adequate informed consent for complex and potentially controversial procedures, use of genetic information in reproductive decision making, and reproductive rights.

Do healthcare personnel properly counsel parents about the risks and limitations of genetic technology?

How reliable and useful is fetal genetic testing?

What are the larger societal issues raised by new reproductive technologies?

Clinical issues including the education of doctors and other health service providers, patients, and the general public in genetic capabilities, scientific limitations, and social risks; and implementation of standards and quality-control measures in testing procedures.

How will genetic tests be evaluated and regulated for accuracy, reliability, and utility? (Currently, there is little regulation at the federal level.)

How do we prepare healthcare professionals for the new genetics?

How do we prepare the public to make informed choices?

How do we as a society balance current scientific limitations and social risk with long-term benefits?

The difference between somatic and germ line gene therapy is transmission (or not) of the transgene to future generations. Somatic cells are all the cells in your body other than those directly involved in reproduction. In the female these are the eggs and in the male it is the sperm and those cells that give rise to sperm (these cells are generically termed gametes). Thus, somatic gene therapy will only effect the individual who receives the treatment since none of the reproductive cells will receive the transgene.

In contrast, gene therapy targeted to the germ cells will result in gametes (eggs or sperm) with the transgene. If the gene therapy patient then has children, the children will also have the transgene.

Most scientists and the public have accepted the idea of somatic gene therapy, it is thought of like any other experimental treatment. There is an ethical and a practical concern that if gene therapy targeted to germ cells is successful then people will try to custom-design their children with enhancing genes that may not be in the best interest for the health or welfare of their children. Abuse of the gene therapy technology targeted to the germ cells is particularly troubling when you realize that modifications to the germ line are permanent, effecting all the future generations

The main area of disagreement is whether human genes themselves can be regarded as inventions or products. The pharmaceutical industry is arguing that they are inventions and that without additional legal protection for gene-based medicines, companies would have no way of getting an adequate return on their investment in DNA research. However, this implies that at present products which use, or are based on, human genes are not protected. This is simply not true as the four main types of medicinal products in this area, small molecule drugs, protein therapeutics, gene therapies and genetic screening tests, are already well-covered by the patent system.

To investigate the variation occurring in the human genome by studying samples collected from populations that are representative of all of the worlds peoples, and

Ultimately, to create a resource for the benefit of all humanity and for the scientific community worldwide. The resource will exist as a collection of biological samples that represents the genetic variation in human populations worldwide and also as an open, long-term, genetic and statistical database on variation in the human species that will accumulate as the biological samples are studied by scientists from around the world.

The Human Genome Diversity Project (HGDP) is working to gather DNA samples from 7 distinct indigenous peoples all over the globe.(1) The presumption is that, if these peoples die out under imperialist oppression, the human race will be no less diverse for the wear.

proposes to collect and catalogue genetic samples from indigenous nations classified as endangered and facing extinction. HGDP will make the samples available to researchers with the goals of discovering these nations histories of migration and evolution and the degree of variance in human genes, and identifying genetic susceptibility to disease

The position of indigenous peoples vis a vis biotechnology is still evolving. The common thread in the various positions is the view that life-forms should not be patented. If the ownership of patents on life-forms is the main incentive for scientists and corporations to invest in biotechnology, it might be a good idea not to allow this. The benevolent motives avowed by scientists who want to contribute to sustainable development should not be tainted by the commercialization or commodification of life.

It should be recognized that indigenous peoples have a right to their intellectual and cultural heritage; This right is being blatantly violated by developments in biotechnology

Customer Analysis

Introduction

When looking at the passenger airline customer, it is necessary to determine the factors that have a significant effect on customer demand for airline travel. To perform this task requires looking at existing demographics of airline customers and the trends of customers of the larger picture.

Method of Analysis

The general focus will be on the US population with some brief reference to international segments and related data. Looking at passengers demand there are two broad factors that are Macro- economic and passenger specific factors. The analysis will focus on those factors that pertain to the latter while the former will be discussed in more detail in the industrial analysis section. The airline passenger can be divided into two groups of, Business and Leisure traveler. These segments are not mutually exclusive as some travel is a combination of both. An over lap of customer may pertain to both groups and therefore represented more than once in data.

Income the Common Factor

Income is viewed as having a strong correlation with passenger travel for both leisure and business travel. While it is harder to segment business travel, as a companys income was generally found that companies bottom line or operating cost were impacted by travel expenses. On a larger scale Gross domestic product was cited as having an impact on business travel.

Leisure travel provides a clearer means of looking at the passengers airline customer. By looking at income as a separate entity segment first allow for an understanding of consumer purchasing power. Based on this data, marketers and statisticians can estimate to greater accuracy, discretionary income for various segments of the population. Through income analysis the industry and markets can streamline their focus of various target markets in the market. Customer trends and potential reactions are masked by their income. Finally looking at customers market segments references are made to their income and spending power or potential spending power.

Income

The Census Bureau has four classifications for income. Based on the alternative definitions only one showed a real decline in median household income from 001 to 00. Money income less taxes declined 0.8% from $7,76 to $7,066. This definition includes money income and realized capital gains less, income and payroll taxes. Money income in the broadest sense as, money received by a person or household during a calendar year, declined 1.1% from $4,00 to $4,40, for the same period. This was the second consecutive annual decline in median household income. Per Capita money income declined by 108% in real terms for the same period to $,74. This represented the first annual decline in per capita income since 11.

Income by Region and Residence

Real media household income did not change for three of the four regions in the US from 001 to 00. The Mid-west was the only region that experienced a reduction of % of median household income at $4,600 for 00. The other regions of the North-east, South, and West maintained their status quo at $45,6, $,5 and $45,14 respectively. Money income for households in the South continued to be the lowest among the regions.

Residence

Median household income in metropolitan areas remained was$ 45,57 down 1.5% representing a second decline consecutively. Households outside of metropolitan remained unchanged at $4,654 in 00. For households inside central cities experienced a decline of 1.% to $6,86; while those outside of central cities recorded a decline of 1.5% to %50,717. Based on a two year average comparison of median households four states in the Mid west experienced declines and three in the South (Florida, Mississippi, North Carolina), in the West (Hawaii, Nevada and Oregon). Marylands household income was higher than 47 States except Alaska and Minnesota, while West Virginia came in lower than 4 states.

Age and Sex

According to the Census Bureau there are 80.5 million men and 71.5 million women working and 7% and 58.6% respectively working full time. Both percentages remain unchanged from 001 to 00. The median earnings for men and women working fulltime year round increased to $,4 from $8,884, and $0,0 from $,680 respectively. The female to male earning ratio was .077 showing no statistical difference however men experienced stagnant or declining earnings from 1. Women on the other hand have not experienced a decline since 15.

Under the age consideration, households under the age of 65 years had a 1.1% decline in median income to $4,510 from $50,010. The age group between 5 and 74 saw a median household decline of .% to $7,88. Similar declines in income were found for other age groups as recorded by the Bureau. Households within the 5 to 65 age boundary had the highest median income ranging upwards of b$50,000 as compared to younger and older age groups.

Income Race

Due to the growing diversity of the US population the Census Bureau saw fit to change or add new classifications to better reflect growth of population. For simplification of the analysis classifications are, white, black, Hispanic, and Asian and pacific islanders.

Median household income for whites fell by 0.6% to $44,64, for blacks .5% to $, for Hispanics .% to $,100 and 4.5% to $54,488 for Asian and pacific islanders. It was observed that for mixed households the decline in median income was greater than the previous numbers. Blacks were reported to have the lowest median income while Asians recorded the highest income for all definition of income.

GROUP SEGMENTATION

The US Affluent Market

For marketing measurement the affluent individual or household falls in the range of

$75K and upwards, annually. Married couples represent 80% of this income segment with 77% of these households having more than one wage earner. Thirty three percent of affluent households have children under the age of 18 years. In todays economy a dual income earning $75K, supporting two children may not have the type of discretionary income or spending habits that would identify them as affluent for marketing purposes. There are 1. million affluent households earning upwards of $100K with 65% in the lowest benchmark between $100 to $14K.

Demographics

The quintessential affluent customer is well educated. Baby boomers comprise the bulk of the affluent households .The age groups of 5-44 and 45-54 account for over half of affluent households. Subtle changes in the affluent population are occurring as ethnic groups are edging upwards in percentage share. Secondly women outnumber men as being affluent representing 54% compared to 46% men. This trend is expected to grow at 4.6% annually. After taxes and other consumer expenditure the average US household has about$1,500 leftover while the typical affluent household has nearly $6K left over to invest or save.

Travel & Trends

For major types of transportation (ships, rented vehicles, trains and airplanes) affluent post spending levels of 144% to 170% above the average spending of households. They represent a large portion of households participating in frequent flyer programs. Market research data by the Simmon group cited affluent as 5% more likely to have taken a trip outside the continental US in the last three years and 75% more likely to have done so by plane. As heavy, representing three or more trips in three years, affluent are heavy foreign, Alaska, or Hawaii travelers. As far as plane travel trips, effluents indices dial in steadily at high levels compared to that of average household travel. These indices increase both for domestic and international travel and the spread increases with annual frequency. About 8% of affluent households take a domestic vacation or personal trip by plane compared to 5% of overall households. This represents 51% above the average household.

Generation Travel

Introduction

In looking at air line travel by generation the study will look at those groups that are potentially significant value to the industry. Over lapping of groups can and will occur when looking at their demographics travel patterns and attitudes. The data however highlights markets that may be neglected or having greater potential to airlines. Generation travel groups the US population in age groups that represents individuals born at a particular time that define the by their cultural experiences of their life.

Active seniors

Demographics

Active seniors are those individuals of adult age between 65-75 years. There are about 18.1 million active seniors in the US (Census Bureau ~7% of overall ) expected to swell to 6 million by 015 and 6 million by 05. In 00 the estimate of active seniors will be approximately 17% of the nations total population. Older Americans are considerably less diverse than their younger counterparts. About 15 million of this group white (84%), 1.4 million black (8%), 1 million Hispanic and less than % Asian.

Active seniors record the highest percentage of discretionary income among any age group. The working mature or pre- retirement group of age 55-64 years also shares a similar position of high discretionary income. An AARP study reported the income of retirees grew faster than younger workers in the past two decades. Between 180 and 1 income among the 50-61 year old, rose 1% while that of active seniors (6-74) rose 7% and 0% for the older group.

Overall active seniors are more likely to be female than male, (65% compared to 44%) with over half living with another person or spouse. The expected aggregate income of this group is expected to reach $. trillion in 006. The average after tax income of this group was $7,567 with a spending average of $,864 representing a full 8.% more than income. By contrast, US households spend 1% of their annual average income of $40,65. The AARP study found that 40% of active seniors fell in the middle and upper income levels and receive pension income. A greater percentage of this group dwells in the Southern and Middle Western regions of the country, with ratios of 5% and 6% respectively. The North East was represented by % of this group.

Travel & Trends

In the US 4% of active seniors have traveled outside the continent in the last three years. The study (AARP) indicates a penetration rate that outstrips those of younger and older age groups. Twenty percent of active seniors traveled by plane outside the US with very little of this travel for business. Some four million or .6% of active seniors took plane trips outside the US in the past three years. Travel loving, seniors take to the air for personal and vacation travel at about the same rates as their younger counterparts.

Factors affecting travel for this group are health, nationality and mindset.

Baby Boomers

Demographics

During the 146-164 period was generally considered to represent the baby boom era where .8-4. million babies were born annually. In 00, the nations 77.6 million Baby Boomers range in age from 8-56 years of age, with 5 million at or over the age of 50.

January 1st 16 stands as the 50 year turning point for this group as will the next 18 years. The Census Bureau estimates that between 15 -050 the over 65 population will more than double. Whites represent the majority of this group at 74% followed by blacks with 11.% and Hispanics.7%. According to Simmon Market research Americans of age 5-54, Boomers are more likely than average to be married represented by 56.6% overall married population. Looking at the Baby Boomer segment separately the ratio jumps to 6.4 % for the group.

Baby Boomers between the ages of 5-54 are 80% employed with 70% employed full time. Based on a projected .% rate the spending power is expected to reach $.7 trillion in the next five years. Estimation was based on a conservative1% increase in annual income of the average after tax income of $54,44 in 00.

The generations greatest expense burdens are financial services, personal insurance and pension. As more boomers edge towards retirement or at least a less active work schedule, they will have more time to take trips. The results are increases in leisure travel will be a windfall to the Travel industry. Data indicate that over 74% of American adults age 5-54 travel within the US last year and over 40%took a foreign trip in the past three years.

Market research indicates five factors that motivate the mature psyche. They are, autonomy/self sufficiency, social and spiritual correctness, altruism, personal growth; and revitalization. Also mentioned for marketing strategy was the groups unwilling to accept aging and stigma associated with Boomers as middle aged or old.

The cities within a city.

If you asked me to tell of a place that was so unique, so incredible, so amazing, then I would only be able to come up with one place. That place is Las Vegas. I can remember becoming incredibly excited whenever I heard that I would be going to Las Vegas, I would get a jolt of energy that would have me bouncing off the walls all day. My family came to call this the "Las Vegas syndrome" because it would be the only time in my shy existence where I would be so excited about something. To me, Vegas wasnt just some place to vacation to, it was a place to be truly alive.

What I love most about Las Vegas is that its not like visiting just one town, its like visiting many towns all rolled into one. Each hotel in Las Vegas has a different theme and with each theme comes a sort of unique culture. You start at the beginning of the Strip and the first thing you see is a giant pyramid with a marvelous sphinx looking down at you beckoning you to enter. If you decide to accept the shpinxs offer youre immediately brought into ancient Egypt complete with the river Nile flowing through the hotel. Hieroglyphics cover the walls, telling "ancient" tales of lucky gamblers who managed to beat the house. Everyone who works there is dressed in some sort of ancient Egypt garb and they treat you as if youre the Pharoh himself. While the inside of the hotel looks like ancient Egypt, the cool air blowing from the air conditioner reminds you that you are still in 1st century. Help with essay on The cities within a city.

Walking down the Strip, for me, is an amazing thing. As gigantic neon signs wage an electric war trying to get me to enter there hotels, I stop and wonder what place I want to visit next. By place I mean which city, or culture do I want to visit next. Do I want to see Rome? Hows New York City doing? Will luck strike me in the Middle East? The hardest thing for me to do in a trip to Vegas would be to decide which hotel I would want to visit, which culture I want to embrace now. Where will I lose myself and enter a sweet dream of being somewhere else.

Exiting the giant pyramid, you are enticed by an equally large castle. This castle has a splendid drawbridge all while surrounded by an empty moat. Entering the castle means entering medieval England and all its splendor. Whimsical music plays in the background cut through by the sounds of joyous laughter as some lucky person hits the big jackpot. Weather jackpots were around during the real medieval days is irrelevant, as long as people win them historical accuracy can be damned. Everything you would expect from a ancient castle town can found here. Noble knights and insane jesters walk through the place, while fair wenches drown your thirst with a cold goblet of ale. Everything has been recreated from the phony accents to the wondrously epic battles between the ever popular blue knight and the infamously despised red knight. If youre lucky the king will bless you with his favor, but unfortunately this does nothing for your odds at the craps tables. The lavish decorations that adorn every inch of the hotel will wow your eyes and make you wish that your other senses could see too. They are just too beautiful to miss, and if you do, youll regret it.

The first time I stepped into Caesars Palace I was awe struck. This is what I imagined ancient Rome to be like, just without all the slot machines and old ladies giving me an evil eye for being to close to there change bucket. The place is truly beautiful. Incredibly well crafted statues of old Roman gods and goddesses are placed throughout the casino eloquently, with my favorite having to be the giant statue of a goddess whose name slips my mind faster than my anniversary. With great marble flooring, lavish paintings, wonderful carpet, and mostly clean air, there is no finer place to stay in Las Vegas. Like the other themed places, all the employees dress like there ancient Roman counterparts. If making you feel like the emperor himself is their job, then by gum they do it awesomely.

Las Vegas is filled with almost any culture imaginable. From the Italians to the Chinese to the Irish, Las Vegas can satisfy your needs to see the world. With wonderfully detailed hotels and authentic service that makes you feel like youre actually at the place that the hotel represents, you are never let down. Sure you could spend all your money and time trying to fly all across the world to visit these places, but why? Las Vegas is like a one stop shop for all the cultures of the world. I could do in about four days something that would take you a month to do if you tried to the hard way. Las Vegas has everywhere you would want to visit all in one area, which makes it a place where no one would ever want to leave because the entire world is just right there waiting for you.
